Bacterial inactivation, DNA damage, and faster ATP degradation induced by ultraviolet disinfection

摘要: • Long amplicon is more effective to test DNA damage induced by UV. • ATP in bacteria does not degrade instantly but does eventually after UV exposure. • After medium pressure UV exposure, ATP degraded faster. The efficacy of ultraviolet (UV) disinfection has been validated in numerous studies by using culture-based methods. However, the discovery of viable but non-culturable bacteria has necessitated the investigation of UV disinfection based on bacterial viability parameters. We used quantitative polymerase chain reaction (qPCR) to investigate DNA damage and evaluated adenosine triphosphate (ATP) to indicate bacterial viability. The results of qPCR effectively showed the DNA damage induced by UV when using longer gene amplicons, in that sufficiently long amplicons of both 16S and gadA indicated that the UV induced DNA damages. The copy concentrations of the long amplicons of 16S and gadA decreased by 2.38 log/mL and 1.88 log/mL, respectively, after exposure to 40 mJ/cm2 low-pressure UV. After UV exposure, the ATP level in the bacteria did not decrease instantly. Instead it decreased gradually at a rate that was positively related to the UV fluence. For low-pressure UV, this rate of decrease was slow, but for medium pressure UV, this rate of decrease was relatively high when the UV fluence reached 40 mJ/cm2. At the same UV fluence, the ATP level in the bacteria decreased at a faster rate after exposure to medium-pressure UV.

摘要: Bacterial adhesion to surfaces and subsequent biofilm formation are a leading cause of chronic infections and biofouling. These processes are highly sensitive to environmental factors and present a challenge to research using traditional approaches with uncontrolled surfaces. Recent advances in materials research and surface engineering have brought exciting opportunities to pattern bacterial cell clusters and to obtain synthetic biofilms with well-controlled cell density and morphology of cell clusters. In this article, we will review the recent achievements in this field and comment on the future directions.

Effects of heavy rainfall on the composition of airborne bacterial communities

摘要: Wet deposition scavenges particles and particle-associated bacteria from the air column, but the impact of raindrops on various surfaces on Earth causes emission of surface-associated bacteria into the air column. Thus, after rainfall, these two mechanisms are expected to cause changes in airborne bacterial community composition (BCC). In this study, aerosol samples were collected at a suburban site in Seoul, Korea before and after three heavy rainfall events in April, May, and July 2011. BCC was investigated by pyrosequencing the 16S rRNA gene in aerosol samples. Interestingly, the relative abundance of non-spore forming operational taxonomic units (OTUs) was always higher in post-rain aerosol samples. In particular, the absolute and relative abundances of airborne always increased after rainfall, whereas those of airborne , including and , consistently decreased. Marine bacterial sequences, which were temporally important in aerosol samples, also decreased after rainfall events. Further, increases in pathogen-like sequences were often observed in post-rain air samples. Rainfall events seemed to affect airborne BCCs by the combined action of the two mechanisms, with potentially adverse effects on human and plant health.

Response of bacterial communities to short-term pyrene exposure in red soil

摘要: Pyrene, a representative polycyclic aromatic hydrocarbon (PAH) compound produced mainly from incomplete combustion of fossil fuels, is hazardous to ecosystem health. However, long-term exposure studies did not detect any significant effects of pyrene on soil microorganism. In this study, short-term microcosm experiments were conducted to identify the immediate effect of pyrene on soil bacterial communities. A freshly-collected pristine red soil was spiked with pyrene at 0, 10, 100, 200, and 500 mg·kg and incubated for one day and seven days. The bacterial communities in the incubated soils were analyzed using 16S rRNA sequencing and terminal restriction fragment length polymorphism (T-RFLP) methods. The results revealed high bacterial diversity in both unspiked and pyrene-spiked soils. Only at the highest pyrene-spiking rate of 500 mg·kg , two minor bacteria groups of the identified 14 most abundant bacteria groups were completely suppressed. Short-term exposure to pyrene resulted in dominance of Proteobacteria in soil, followed by Acidobacteria, Firmutes, and Bacteroidetes. Our findings showed that bacterial community structure did respond to the presence of pyrene but recovered rapidly from the perturbation. The intensity of impact and the rate of recovery showed some pyrene dosage-dependent trends. Our results revealed that different levels of pyrene may affect the bacterial community structure by suppressing or selecting certain groups of bacteria. It was also found that the bacterial community was most susceptible to pyrene within one day of the chemical addition.

Spatial and seasonal variations in bacterial communities of the Yellow Sea by T-RFLP analysis

摘要: Four typical coastal sites (rocky shore, sandy shore, mud flat shore, and artificial harbor) at the Yellow Sea were chosen to investigate the spatial and seasonal variations in bacterial communities. This was accomplished by using terminal restriction fragment length polymorphism (T-RFLP) analysis of PCR amplified 16S rDNA fragments. Two kinds of tetrameric restriction enzymes, I and I, were used in the experiment to depict the bacterial community diversity in different marine environments. It was found that the community compositions digested by the two enzymes separately were different. However, the results of bacterial community diversity derived from them were similar. The MDA analysis results of T-RFLP profiles coming from I and I both exhibited a significant seasonal community shift for bacteria and a relatively low spatial variation among the four locations. With I as the sample, the pair wise -tests also revealed that variations were minor between each pair of marine environments, with ranging from 0.198 to 0.349. However, the bacterial community structure in the mud flat site depicted a larger difference than each of the other three sites ( ranging from 0.282 to 0.349).
